Background: Colorectal cancer (CRC) is the third most common malignancy and the second leading cause of cancer-related death throughout the globe. Therefore, there is an imperative demand to find effective therapeutic options for controlling of this disease. Ajwa dates are famous for their high contents of phytochemicals that possess nutritious values and numerous medical benefits including fighting cancers.
Aim: this study aimed to determine the total polyphenol and flavonoid contents in Ajwa dates and to investigate an impact of Ajwa date extracts on Human colorectal cancer cells (HCT116).
Methods: Chloroform, ethyl acetate and butanol extracts prepared from Ajwa date were prepared by solvent-solvent fractionation. The viability of HCT116 cells treated with the extracts was assessed by MTT assay. Effect of the extracts on the clonogenicity and cellular migration potential of the HCT116 were also examined. Morphological changes were observed by Giemsa staining. Agarose gel electrophoresis and comet assays were performed to assess the pro-apoptotic potential of the extract. Finally, levels of the reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) were determined by DCFH-DA and JC-1 dyes, respectively.
Result: Treatment of HCT116 cells with the chloroform, ethyl acetate or butanol extract indicated the Ajwa Date Butanol Extract (ADBE) was the most effective fraction. It markedly suppressed the proliferation, colony formation and cellular migration potentials of the HCT116 cells. The IC50, after 24, 48 and 72 h of treatments were 250, 187.5 and 125 μg/100 μl, respectively. ADBE-treated HCT116 cells displayed morphologic and biochemical features of apoptotic cell death. The ADBE also increased level of the intracellular ROS and induced permeabilization of the MMP.
Conclusion: These findings provide valuable insights on the bioavailability of ADBE as a promising agent for development innovative medicines against colon cancer.
