The Protective Effect of Hypericum revolotum Extract against Changed in Vascular Reactivity in Metabolic Syndrome in Rats

Abstract : Aim: The study was conducted to evaluate the efficacy of Hypericum Revolotum total extract (HRT), its active fraction and its isolated compounds in protecting metabolic syndrome rats from vascular dysfunction. Methods: Male albino rats (n=52) weighing 180-200 g were randomly divided in to two groups; control and MetS. MetS was induced by adding 10% fructose in drinking water and 3% salt in diet for 12 weeks. The in-vitro effect of the natural compounds on vascular reactivity was studied through incubation with the isolated thoracic aorta from MetS animals. Vascular reactivity was studied by the contraction and relaxation response of the isolated aorta to phenylephrine (PE) and acetylcholine (ACh) respectively. The direct relaxation effect of the compounds on PE pre-contracted aorta was also studied after incubation with Nω-nitro-l-arginine methyl ester hydrochloride (L-NAME), Wartmannin, (TEA), indomethacin, methyline blue (MB), potassium chloride (KCl) and Atropine. Furthermore, vascular nitric oxide (NO) release and the in vitro effect of the compounds on advanced glycation end-products (AGEs) formation was also studied. Results: HRT, some active fractions and its isolated compounds significantly alleviated the exaggerated vasoconstriction and impaired vasorelaxation in MetS aorta. They showed a direct vasodilation effect in pre- contracted aorta which was completely blocked by L-NAME and partially inhibited by Wartmannin and TEA), HRT, the active fraction (Fr.3) and its isolated compounds significantly stimulated the release of aortic NO in a dose dependent manner and reduced AGEs formation. Conclusion: HRT, some active fractions and its isolated compounds have protective properties against vascular dysfunction associated with MetS or AGEs possibly through stimulating the release of endothelial NO and reducing AGEs formation.
