Antagonistic activity of some actinomycetes isolated from different environments against some human pathogens

Abstract : Eighty one actinomycete isolates were isolated from rhizosphere soil, mine soil, fresh and marine water, shrimp peel and plant leaves and roots from different localities of Jeddah and Al-Madinah Al-Munawwarah, Saudi Arabia. All isolates were screened for antimicrobial activity on Muller-Hinton using well diffusion method .Nine actinomycetes isolate which exhibited antimicrobial activity. Six isolates were showed active against human pathogenic bacteria and Candida albicans. Only three isolates were highly activity against some fungi. The isolate D8 was the most active isolate against some human pathogenic bacteria including multiple antibiotic resistant Methicillin-resistant Staphylococcus aureus (MRSA), Vancomycin-resistant Enterococcus ,Acinetobacter baumannii, Staphylococcus aureus ATCC 29213, Streptococcus pyogenes, Escherichia coli ATCC 35218, Salmonella typhimurium and Shigella sp. in addition to Candida albicans. The isolate D8 was characterized and identified through physiological, biochemical tests and culture characteristics in different culture media, in addition to examination with light and scanning electron microscope as Streptomyces sp. The maximum antimicrobial activity of the selected isolate D8 was achieved using Starch nitrate medium at pH 8 and after 5 days of incubation at 28oC. The antimicrobial agent was extracted using Ethyl acetate (1:1) and purified using bioautography. The active material affected the growth, morphology, cells number and cell wall composition of Gram-positive and negative bacteria. The active material enhanced cell permeability of K+ and increased cell respiration.
